What is Poly-L-Lysine used for?
Poly-L-Lysine is used to coat tissue culture plasticware for enhanced cell attachment and adhesion. Coated surfaces will often improve cell attachment in reduced or serum-free conditions. The optimal concentration for cell attachment and culture may differ for various cell types.
What is the difference between Poly D lysine and Poly-L-Lysine?
Poly-D-lysine refers to a type of polylysine produced by the polymerization of D-lysine while poly-L-lysine refers to a type of polylysine produced by the polymerization of L-lysine. Thus, this is the main difference between poly D lysine and poly L lysine.
Is Poly-L-Lysine toxic to cells?
The combination of two differently charged polypeptides, poly-L-lysine (PL) and poly-L-glutamate (PG), has shown excellent postsurgical antiadhesive properties. However, the high molecular, positively charged PL is toxic in high doses, proposed as lysis of red blood cells.
How do you use Poly-L-Lysine for coverslips?
- Place glass coverslips in 6-well dish using special forceps for coverslips.
- Add 2 ml of poly-L-lysine (0.1% from Sigma, diluted 1:10 in sterile water) to each well.
- Wash 3x with sterile PBS.
- Place under the hood UV light 10-15 min to sterilize (remove cover).
- Plate cells as usual (use 2-2.5 ml for each well).
How long does Poly lysine last?
Remove poly-d-lysine solution and rinse with sterile PBS or water 1-2X. Do not scrape the bottom of the coated plates or flasks. Use plates immediately or store with PBS for up to 5 days.
What is poly L-lysine coating?
DESCRIPTION. Poly-L-Lysine is a synthetic amino acid chain that is positively charged and widely used as a coating to enhance cell attachment and adhesion to both plasticware and glass surfaces. Certain cell types secrete proteases, which can digest Poly-L-Lysine.

How do you wash coverslips?
Wash the coverslips extensively in distilled water (two times), then double distilled water (two times). Be sure to wash out the acid between sticked coverslips. Rinse coverslips in 100% ethanol and dry them between sheets of whatman filter paper. Keep coverslip in a clean container.
How do you treat a Poly-L-Lysine slide?
Coating Procedure:
- Allow diluted Poly-L-Lysine solution to come to room temperature (18-26°C) before use.
- Place clean slides, a rack at a time, in diluted Poly-L-Lysine solution for 5 minutes.
- Drain slides and dry in 60°C oven for one hour or at room temperature (18-26°C) overnight.

How do you store Poly-D-lysine plates?
Storage: Store coated plates at 4-8* or room temperature. Store PDL solution at 2-8* for shorter periods of time. Store at -20* for extended storage times.
